wonder drug-Mestinon- is loosing its effect-Why?

Started on Mestinon about 9 months ago by my family doctor. Results were amazing for several months. I was able to return to all normal activities. Over the last couple months I have been having to increase the frequency and then the dose ( from 60mg three times daily to 75mg four times daily). As all tests were initially negative, except for my positive response to Mestinon, I am unable to receive any other treatments. I have two main questions. Does the effect of Mestinon decrease with time? I have really been having to take it easy; any yard work, painting etc. wipes me out for 24-48 hours even though I'm on this medication. Secondly, is it possible to have once negative blood work and emg results become positive over time; should I ask to be retested? I am definitely worse without Mestinon, but it's once amazing effects seem to be decreasing.
